Transport Scotland has said the A77 will be closed between Dutchhouse roundabout at Monkton to the Bellfield roundabout this weekend.
The closure is to facilitate part of the £10.6m improvement scheme on the key arterial route between Glasgow and Stranraer.
The road will be closed from 8pm on Saturday (12 October) until 9am on Sunday 13 October to allow two new flyovers to be lifted into place.
Diversions along the A71 and A78 will be in operation for the duration of the closure.
The authority's Project Manager for the A77 Symington-Bogend Toll scheme, Ruairidh Campbell, said: "The two new flyovers over the A77 carriageway are expected to be lifted overnight this Saturday.
"They form part of a wider improvement scheme on this section of the A77, which will see improved safety and accessibility as well as minimise delays on the route.
"The lifting process and placement of the flyers will no doubt be a spectacular but challenging feat, and to protect drivers' safety we will be required to close the road between Dutchhouse and Bellfield roundabouts for the duration of the work.
"As there will be no access to this section of the A77 to road users, I would ask all those who use the route to be aware of the diversion routes and plan their journeys accordingly."
The £10.6m A77 Symington-Bogend Toll improvement project, which will see the central reservations between Spittalhill Interchange and Dutch House roundabout closed, and the two new flyover bridges replacing the need for right hand turns across the busy stretch of road.
Contractor RJ McLeod is around mid-way through construction.
The project is scheduled for completion in summer 2014.
